Trisha, Oliver's girlfriend, had sex with Ben, his brother. Lydia and Regina are the mother and the sister of the two boys. :)

REGINA What is the big deal?

LYDIA This is horrible. That Trisha girl is pregnant.

REGINA Do we know who the father is?

LYDIA Given the way she’s gone through this family like pink-eye, it’s kind of up in the air, isn’t it?

Pink eye is congiuntivite, according to the English-Italian dictionary. Lydia is comparing Trisha to pink eye because it can spread easily from one person to another and Trisha has gone from one person in the family to another. When Lydia says "up in the air" she means that it is unclear who the father is.

Jacob has summed things up perfectly...just to add "Pink eye" is a common contagious eye infection that refers to the redness and the irritation that shows up in the eye as a result.
