Dean and Sam are watching security footage showing a killer, when Sam notices a bright flare in his eyes. Dean:
-- Well, maybe it’s just a camera flare.
-- That’s not like any camera flare I’ve ever seen. You know, a lot of cultures believe that a photograph can catch a glimpse of the soul.

The definite article is used because it's the soul of a person. A person has only one soul, hence THE. It's not generic.

Am I right? Thank you.
