Is this correct if I say: "I need a little bit of a room" in order to express that I need more space? Can I use it in the situation when I'm crowded by people and I'd like to make some more space around me?

No. "I need a little bit of a room" means that you need a small room. If you need more space, you "need a little bit of room."
