- Do you know where my book is? I've looked everywhere, I can't locate it.

- "I can't locate it" here means "I can't find it anywhere", right?

    Unknoewn is right, of course.

    But it's a strange word to use (too formal). Maybe it should be either:

    "Have you seen my book lately? I have searched everywhere and can't locate it."

    or

    "Do you know where my book is? I've looked everywhere and still can't find it"

    EDIT: I meant it's too formal for the context (the rest of the sentence) :D
