Hopefully a quick query. When the answer to a question is:

No it's not or yes it is

Would it be better to have a comma after the no and yes, so it reads:

No, it's not/Yes, it is.

Also, would the word 'unfortunately' more than always be followed by a comma?

The commas after Yes and No are needed; and you are correct about "unfortunately" often taking a comma -- but not when used adjectivally, as in: "He was unfortunately killed in an auto accident."

With regards to the word unfortunately, would you say that applied for the word eventually as well.
Please see the following sentence.

He ran as fast as he could for as long as he could, then eventually, he gave up.

Seems correct to m;, but the comma after "could" should be changed to a semicolon inasmuch as "then" acts as a conjunction between two complete sentence parts (each with its own subject and verb), in one of which there is already a comma. I think some people would recommend an additional comma after "then," but I consider it optional.
