How to put Tashkil in the word روحه (his soul)؟
I heard in the past that one can't put 2 sukuns consecutively, I'm not sure about this case if I can consider رُوْحُهُ as correct.

That's only 1 sukoon consecutively in روحه. The only places I've seen 2 sukoons used back to back, is in spoken Arabic. But that is still extremely rare...from my observation.

That said; the way you spelled روحه is correct.

You aren't supposed to put Tashkil on the letter preceding حرف علّة or حرف علّة itself. The rule is حروف العلة لا تقبل الحركات.
As for the Tashkil for the last letter in the word, it depends on the context, and more specifically إعراب.

    Here are the basics:
    • If the word روحه is a subject (i.e., فاعل), the last letter should be Dammi or Tanween Madmom (i.e., مرفوع). Example: إنفَصَلَت روحُهُ عن جَسَدِه
    • If the word روحه is an object (i.e., مفعول به), the last letter should be Fatha or Tanween Maftoh (i.e., منصوب). Example: أهداكَ اللّهُ روحًا وريحانًا
    • If the word روحه comes after a preposition (i.e., اسم مجرور) the last letter should be Kasra or Tanween Maksor (i.e., مجرور). Example: تخلّى جَسَدُهُ عن روحِهِ
    I hope this helps.

So in this case I have to choose
    قد يمسك بممتلكاته أو روحِهِ?
(Not sure if أو is considered a preposition)

Yes. In the example you gave "أو" is considered حرف عطف (a coordinating conjunction). The general rule in this case is the word before it and after it get the same Tashkil. The way you wrote it is correct.
