    Hello,
    would it be idiomatic to say in a friendly environment when i am inviting the addressees to ask the questions?

    'I will happily assist you by replying your questions.'

    Would the part 'replying your question' have to be introduced by a preposition 'by'?

    Yes, you need "by." You also need to say "replying to your questions." "Reply" is not a synonym for "answer."
