Since "leading" isn't defined and could mean any number of things (greatest volume, highest survival, best rating in some magazine survey, etc.), I think that an equally ambiguous word such as "importante" is faithful to the original.

Based on the context you give us, I would go with ... "Nuestro hospital es el centro de traumatologia líder en xxx lugar," since the one who leads is the one who goes in front of others, in advance of others, it is the first.
I have seen that in different magazines and newspapers in Spanish and the idea is the same as in the statement you gave us. Ex. " XYZ, el hospital líder en el tratamiento de ..." etc. etc.
I also like "El más destacado."

Personalmente me gusta tu propuesta inicial para traducir "the leading"; el más importante (el más destacado; el más relevante; el principal). Personalmente también diría centro traumatológico antes que de traumatología, pero esto es ya cuestión de gustos;
Nuestro hospital es el centro traumatológico más importante de X.
De lectura un poco más libre, tal vez también, por ejemplo;
Nuestro hospital cuenta con el servicio de traumatología más importante en X.

Just my two cents: Thinking about how hospitals market themselves, at least in English, "most important" sounds a little bit unusual to me.
The hospital usually wants to be thought of as the best, the most outstanding, the top trauma facility in a certain area. To me, "leading" is just a slightly more professional way of saying "We're the best!" Best in what way? We don't know. This kind of marketing often leaves out the fine details. But I just get the sense that they aren't trying to say "We're the most important!"

Aquí caben diversas opciones dependiendo de dónde quiera ponerse el acento (más importante, principal; más prestigioso, más destacado; número uno; etc.). "De referencia" creo que incluye connotaciones no implícitas en el original, pero casi cualquier opción sería válida a la hora de traducir leading en este caso. La cuestión es intuir cuál es la que mejor se ajusta al mensaje que quiere transmitir la propia institución en este caso.

Interesting opinions, all of them valid and useful. Marketing is not part of this mix; the hospital exists in a national health structure, but I agree that this is a way of saying "the best" without offending docs at other facilities. Why this quote arrived in English instead of Spanish in the first place is a mystery to me, frankly, but I asked for "the Spanish original" and was told that there was none. I appreciate your help. I'll let you know what ends up in print about the second week in June. Abrazos.

Por acá pasa lo mismo que indicas, nadie quiere decir que es "el mejor hospital/centro/etc." para no ofender a las demás instituciones.
En Puerto Rico, y para propósitos de mercadeo, se suele decir: el Hospital XYZ es uno de los principales centros de trauma de la isla .... la Clínica ABC es uno de los principales hospitales oncológicos del Caribe

Me parece una lectura perfectamente natural.
Aunque no es lo mismo "a leading trauma facility" (uno de los principales centros traumatológicos; se asume que hay otros centros equivalentes en importancia/calidad a este) que "the leading trauma facility" (donde se asume una clara superioridad sobre el resto, por el motivo que sea).
