    Yes, Leider haben Sie verloren or Sie haben leider verloren. In either case: no comma.

    No comma, but usually an exclamation mark:
    "Sie haben verloren!"
    A dot is possible, too.
    "Sie haben verloren."

    Without exclamation mark I would suppose "They lost, unfortunately."
    In case of "Leider haben Sie verloren." it is clear.
    The exclamation mark intensifies that you are not really unhappy that you have won the game.
