Dear friends,

a. What's the population in the US?
b. How many people are there in the US?

- Which sentence is more natural?

Thanks a lot!

Both might be said (after the correction to "of"). I think a) sounds usual, and b) slightly colloquial. In fact, b) is potentially not the same as a) in that you might count holiday-makers and other itinerants you may not wish to count as "population".
