To replace, to substitute?

Which would be the best verb to use, if I wanted to say that someone replaced someone else at work, because the other person was temporarily absent (either because of illness or maternity leave or anything else).

Does it sound ok if you say Mrs. J was replacing Mrs. L?

Step in for someone seems a bit too informal. It should be a neutral word.
